In this study, we characterize the role of the O-antigen, a fundamental part of the bacterial pathogen Pseudomonas aeruginosa. The O-antigen is a polymer of sugar molecules which extends from the surface of the bacteria and into the environment in which it resides. Here we have devised a method to genetically engineer bacteria to express different types of O-antigen with focus on two specific serotypes: O4 and O12. We proceed to study how this affects the bacteria's ability to adhere to plastics, how it affects specific outer membrane proteins such as the type IV pili, and finally how it affects the bacteria's ability to cause infection.
